Ces mol\u00e9cules constituent une cause fr\u00e9quente d\u2019intoxication chez le chien.<br \/>\nOn distingue deux grandes cat\u00e9gories de rodenticides anticoagulants selon leur puissance et leur dur\u00e9e d\u2019action : les rodenticides anticoagulants de premi\u00e8re g\u00e9n\u00e9ration et ceux de seconde g\u00e9n\u00e9ration.<br \/>\nLe warfarine fut le premier rodenticide anticoagulant utilis\u00e9, jusqu\u2019\u00e0 ce que les rongeurs d\u00e9veloppent une r\u00e9sistance, entra\u00eenant la mise sur le march\u00e9 des ARs de seconde g\u00e9n\u00e9ration. Ces derniers (ex. : brodifacoum, bromadiolone, dif\u00e9thialone) sont plus puissants et ont une action prolong\u00e9e par rapport aux mol\u00e9cules de premi\u00e8re g\u00e9n\u00e9ration.<br \/>\nDans une \u00e9tude r\u00e9cente (Paulin et al., 2024), la majorit\u00e9 des chiens intoxiqu\u00e9s avaient ing\u00e9r\u00e9 un rodenticide anticoagulant de seconde g\u00e9n\u00e9ration.<\/p>\n<p><strong>Comment agissent les rodenticides anticoagulants ?<\/strong><\/p>\n<p>Apr\u00e8s ingestion, les ARs sont absorb\u00e9s au niveau intestinal en quelques heures. Les m\u00e9tabolites sont ensuite transport\u00e9s jusqu\u2019au foie, o\u00f9 ils inhibent l\u2019enzyme vitamine K1 \u00e9poxyde r\u00e9ductase. Cette enzyme est indispensable \u00e0 la r\u00e9g\u00e9n\u00e9ration de la vitamine K1 sous sa forme active (KH2, Figure 1), cofacteur essentiel \u00e0 la synth\u00e8se et l\u2019activation de plusieurs facteurs de coagulation (II, VII, IX et X).<br \/>\nLes rodenticides anticoagulants entra\u00eenent donc une d\u00e9pl\u00e9tion de la vitamine K1 active, induisant une baisse progressive des concentrations plasmatiques des facteurs de coagulation concern\u00e9s. Cela conduit \u00e0 un \u00e9tat d\u2019hypocoagulabilit\u00e9 pouvant engendrer des h\u00e9morragies spontan\u00e9es.<\/p>\n<p><strong><em>Figure 1.<\/em><\/strong><em> Sch\u00e9ma de l\u2019action des rodenticides anticoagulants d\u00e9riv\u00e9s de la warfarine : dans l\u2019h\u00e9patocyte, les facteurs vitamine K-d\u00e9pendants doivent \u00eatre carboxyl\u00e9s pour devenir actifs. Ce processus biochimique n\u00e9cessite la forme r\u00e9duite de la vitamine K, qui est oxyd\u00e9e lors de la r\u00e9action. L\u2019enzyme \u00e9poxyde-r\u00e9ductase est indispensable pour recycler cette vitamine et la rendre de nouveau disponible. Les ARs bloquent l\u2019action de cette enzyme, emp\u00eachant l\u2019activation des facteurs II, VII, IX et X, qui s\u2019accumulent alors sous forme inactive. L\u2019administration de vitamine K1 permet de r\u00e9tablir rapidement la situation en r\u00e9activant ce processus.<\/em><\/p>\n<p>&nbsp;<\/p>\n<p><img loading=\"lazy\" decoding=\"async\" class=\"aligncenter wp-image-31465 size-full\" src=\"https:\/\/www.mylav.net\/wp-content\/uploads\/2025\/04\/img1.png\" alt=\"\" width=\"640\" height=\"358\" \/><\/p>\n<p>&nbsp;<\/p>\n<p><strong>Quels signes cliniques peuvent appara\u00eetre apr\u00e8s ingestion ?<\/strong><\/p>\n<p>Les sympt\u00f4mes apparaissent g\u00e9n\u00e9ralement entre 2 et 5 jours apr\u00e8s l\u2019ingestion, en lien direct avec l\u2019\u00e9tat d\u2019hypocoagulabilit\u00e9.<br \/>\nLa cavit\u00e9 pleurale est le site d\u2019h\u00e9morragie le plus fr\u00e9quent. D&rsquo;autres localisations incluent, par ordre de fr\u00e9quence : la peau et le tissu sous-cutan\u00e9 (ecchymoses), les voies respiratoires (y compris le parenchyme pulmonaire), la cavit\u00e9 buccale, le tractus gastro-intestinal, la cavit\u00e9 p\u00e9riton\u00e9ale, le m\u00e9diastin, le syst\u00e8me urog\u00e9nital, la conjonctive et la scl\u00e8re (Figure 2).<br \/>\nLes <strong>signes cliniques<\/strong> les plus fr\u00e9quents sont : l\u00e9thargie, dyspn\u00e9e, anorexie, p\u00e2leur des muqueuses, toux, intol\u00e9rance \u00e0 l\u2019effort et h\u00e9moptysie. On peut \u00e9galement observer : \u00e9pistaxis, saignements gingivaux, m\u00e9l\u00e6na, h\u00e9maturie, p\u00e9t\u00e9chies, ecchymoses et signes neurologiques (convulsions, parapar\u00e9sie).<\/p>\n<p><strong><em>Figure 2.<\/em><\/strong><em> Localisation des saignements chez 81 chiens intoxiqu\u00e9s par des rodenticides anticoagulants. D\u2019apr\u00e8s Paulin et al., 2024.<\/em><\/p>\n<p>&nbsp;<\/p>\n<p><img loading=\"lazy\" decoding=\"async\" class=\"size-full wp-image-31472 aligncenter\" src=\"https:\/\/www.mylav.net\/wp-content\/uploads\/2025\/04\/img2.png\" alt=\"\" width=\"606\" height=\"248\" \/><\/p>\n<p>&nbsp;<\/p>\n<p><strong>Comment diagnostiquer une intoxication aux rodenticides anticoagulants ?<\/strong><\/p>\n<p>Le diagnostic repose sur l\u2019exclusion d\u2019autres causes d\u2019hypocoagulabilit\u00e9 et sur l\u2019anamn\u00e8se. Une enqu\u00eate minutieuse est indispensable pour identifier une ingestion accidentelle ou intentionnelle de substances toxiques.<br \/>\nL\u2019\u00e9valuation du profil de coagulation est essentielle, via le <strong>temps de Quick = PT (= Temps de Prothrombine)<\/strong>qui explore la voie extrins\u00e8que, et le <strong>temps de c\u00e9phaline activ\u00e9e = aPTT (=TCA)<\/strong>, qui explore la voie intrins\u00e8que.<br \/>\nParmi les facteurs de coagulation d\u00e9pendants de la vitamine K (II, VII, IX, X), le facteur VII a la demi-vie la plus courte. Ainsi, une intoxication par AR entra\u00eene d\u2019abord un allongement du PT avant le TCA. Le PT est donc le param\u00e8tre le plus sensible pour d\u00e9tecter pr\u00e9cocement une intoxication.<br \/>\nUne normalisation rapide du PT apr\u00e8s administration parent\u00e9rale de vitamine K1 est un indicateur diagnostique fort.<br \/>\nLa confirmation peut se faire par dosage des concentrations d\u2019AR dans le s\u00e9rum ou le contenu gastrique, ou par analyse toxicologique post-mortem sur foie ou rein (chromatographie liquide).<\/p>\n<p><strong>Quel traitement mettre en place ?<\/strong><\/p>\n<p>Le traitement repose sur l\u2019administration de vitamine K1. Avant de l\u2019administrer, il est important d\u2019\u00e9carter d\u2019autres causes d\u2019hypocoagulabilit\u00e9, car m\u00eame une seule injection peut fausser les r\u00e9sultats des tests de coagulation.<br \/>\nLa dur\u00e9e du traitement d\u00e9pend de la dose et du type d\u2019AR ing\u00e9r\u00e9. Cependant, comme ces \u00e9l\u00e9ments sont rarement connus, on recommande une administration de <strong>vitamine K1 \u00e0 2,5\u20135 mg\/kg\/jour en 2 prises<\/strong>, pendant <strong>au moins 4 semaines<\/strong>.<br \/>\nEn cas de signes cliniques s\u00e9v\u00e8res, une hospitalisation en soins intensifs peut \u00eatre n\u00e9cessaire.
